Establishment of High Efficiency Regeneration System of Alfalfa

摘要 以黑龙江主栽苜蓿品种肇东苜蓿、敖汉苜蓿、公农1号苜蓿、公农2号苜蓿为受体材料,分别研究了外植体类型对不定芽分化的影响,基因型、PGRs的不同配比对子叶节不定芽分化的影响,并确定了芽分化、芽伸长及再生植株生根时的最佳培养基。结果表明,当BA浓度为1.0mg/L时最适于子叶节的分化;当BA浓度为0.5mg/L时植株生长最佳。芽分化培养基为:MS+1.0mg/L BA,30g/L蔗糖,0.8%琼脂,pH5.8;芽伸长培养基为:MS+0.5mg/LBA,30g/L蔗糖,0.8%琼脂,pH5.8;再生植株生根培养基为:MS+1.5mg/L IBA,15g/L蔗糖,0.8%琼脂,pH5.8。 The effects of different explant on buds differentiation,PGRs on adventitious buds regeneration of cotyledonary nodes and factors on regenerated plant of "Zhaodong', "Aohan" ,"Gongnong No. 1" and "Gongnong No. 2" were studies. The result indicated:BA 1.0mg/L was the best;When BA was 0. 5mg/L, the plant developed in good state. The optimal medium for root generation was MS+ 1.5mg/L IBA, 15g/L Sucrose,0. 8%Agar,pHS. 8.
