摘要
本试验旨在探讨顺9,反11-共轭亚油酸对离体培养猪肝细胞、结肠粘膜上皮细胞增殖、PGE2分泌和PPAR-γ基因表达的影响。试验分5个处理,对照组为DMEM/F12培养液,其他处理分别在对照组的基础上添加100μmol/L亚油酸、25、50、100μmol/L顺9,反11-共轭亚油酸;每个处理设6个重复(PPAR-γmRNA定量测定4个重复)。细胞用血清培养24h后处理,继续培养72h。检测上清PGE2分泌、细胞增殖和细胞PPAR-γmRNA拷贝数。结果表明培养液中添加顺9,反11-共轭亚油酸对肝细胞增殖和PGE2分泌都有促进作用(P<0.05),但对PPAR-γ基因表达没有影响;培养液中添加顺9,反11-共轭亚油酸对结肠粘膜上皮细胞增殖影响不显著(P>0.05),对PGE2分泌有显著的促进作用(P<0.05),25μmol/L顺9,反11-共轭亚油酸组PPAR-γmRNA拷贝数显著高于对照组(P<0.05),100μmol/L顺9,反11-共轭亚油酸组极显著高于对照组(P<0.01)。可见,在离体培养条件下,顺9,反11-共轭亚油酸对猪肝细胞和结肠粘膜上皮细胞PGE2分泌都有促进作用,只对结肠粘膜上皮细胞PPAR-γ基因表达有促进作用,这显示顺9,反11-共轭亚油酸对PPAR-γ基因表达的调控可能具有细胞类型的特异性。
This research is aimed to investigate the effects of cis9, trans11-conjugated linoleic acid (c9, t11-CLA) on proliferation, PGE2 secretion and PPAR-γ gene expression of pig hepatocyte and colon mucosa cell cultured in vitro. Five treatments,which were DMEM/F12 (control), DMEM/F12 + 100 μmol/L linoleic acid, DMEM/F12 + 25μmol/L c9, t11-CLA, DMEM/F12 + 50 μmol/L c9, t11-CLA and DMEM/F12 + 100 μmol/L c9, t11-CLA, with six replications each (four replications for PPAR-γ mRNA detection), were designed for the experiment. Pig hepatocytes and colon mucosa cells were cultured for 48 h, and then treated with linoleic acid or c9, t11-CLA for 72 h. Cell proliferation, PGE2 secretion and PPAR-γ, mRNA were detected. The results showed 1) All the treatments of c9, t11-CLA improved cell proliferation and PGE2 secretion of pig hepatocyte (P〈0.05), but no significant was found in PPAR-γ, mRNA copies;2) All the treatments of c9, t11- CLA improved PGE2 secretion significantly (P〈0.05), but had no significant difference on proliferation of pig colon mucosa cell cultured in vitro, and 25μmol/L c9, t11-CLA improved PPAR-γ, mRNA copies significantly (P〈0.05), 100 μmol/L c9, t11-CLA very significantly (P〈0.01). C9, t11-CLA could improve PPAR-γ, gene expression of pig colon mucosa cell cultured in vitro ,which indicated that the activities of c9, t11-CLA on regulating PPAR-γ, gene expression might have the cell type specificity.
