铁对泌乳期小鼠乳腺中乳铁蛋白基因mRNA水平的影响

摘要 探讨铁对小鼠乳腺中乳铁蛋白(lactoferrin,LF)mRNA基因表达的影响。选取雌性昆明白小鼠40只,随机分成5组,按实验动物标准饲养,各处理组添加不同水平和来源的铁:硫酸亚铁(Fe0mg/kg)、硫酸亚铁(Fe60mg/kg)、硫酸亚铁(Fe120mg/kg)、甘氨酸铁(Fe120mg/kg)、甘氨酸铁+硫酸亚铁(Fe120mg/kg)。配种、产仔,于哺乳21d后处死取样,用相对定量RT-PCR方法,以β-actin作内标,测乳腺LFmRNA丰度。试验表明,不同来源和剂量的铁可不同程度影响LFmRNA的表达,甘氨酸铁(Fe120mg/kg)组较对照组差异达极显著水平(P<0.01)。因此,甘氨酸铁可显著提高乳腺中LFmRNA的表达量,作为有效铁补充剂的同时可能促进乳汁中LF的含量。 To investigate the effects of iron on LF (Lactoferrin) gene expression of mice mammary glad. Forty female Kunming white mice were selected and divided into 5 groups randomly. Feeded them according experimental animal standard and add iron of different source and dose: control(Fe 0 mg/kg), FeSO4 (Fe 60 mg/kg), FeSO4 (Fe 120 mg/kg), Fe-Gly(Fe 120 mg/kg),Fe-Gly + FeSO4 (Fe 120 mg/kg). After hybridize and litter, at the twenty-first day after lactation killed all of the mice and collected sample immediately. For investigating LF mRNA expression with relative quantitative RT-PCR using β-actin as an internal standard. The results showed that different source and dose of iron can affect the expression of LF mRNA, especially Fe-Gly (Fe 120 mg/kg) can increase LF mRNA expression significantly(P〈0.01). The result suggest iron is a important factor affect LF mRNA expression level, as a iron supplement Fe-Gly may be increased the level of LF in milk.
