摘要 目的研究柯萨奇B组3型病毒MKP株结构蛋白VP1基因序列及变异性。方法在Hela细胞中增殖病毒,应用RT-PCR扩增目的基因片段,与pMD18-T载体连接,鉴定后测序,进行序列同源性及系统发生分析。结果CVB3/MKP株VP1基因含930个碱基,编码310个氨基酸,与其他参考株比,核苷酸同源性在82.50%以上,氨基酸同源性在95.47%以上,CVB3/MKP株VP1基因与Nancy株聚簇。结论CVB3/MKP株VP1基因与Nancy株在进化上属同一分支。CVB3/MKP株与CVB3/P株在系统发生树中的距离最近。 Objective To investigate the sequence and variation of VP1 gene of a strain coxsackievirus B3 (CVB3)/MKP. Methods The virus was cultured in HeLa cells. Target gene fragments were amplified by RT-PCR and then cloned to plasmid pMD18-T vectors connected to the plasmid to determine sequence after identification and to analyze the sequence homology and phylogenesis. Results The VP1 gene of strain CVB3/MKP nucleotides contained 930 basyls, encoding 310 amino acids, had more than 82. 50% of nucleotides homology and more than 95.47% of amino acids homology, compared with other reference strains. CVB3/MKP VP1 gene clustered with Nancy strain. Conclusions The VP1 gene of strain CVB3/MKP and Nancy strain belong the same evolutional branch and there may be a close evolutional relationship between strain CVB3/MKP and CVB3/P.
