染料木黄酮对大鼠移植动脉血管平滑肌细胞的影响 被引量:2

Effect of genistein on vascular smooth muscle celts in arotic transplants of rats

摘要 目的 研究染料木黄酮(Gen)对大鼠移植动脉血管平滑肌细胞的影响。方法 以BN大鼠为供者,Lewis大鼠为受者,建立腹主动脉移植模型,将受者随机分为3组。实验组(n=8):受者术后腹腔注射Gen20mg·kg^-1·d^-1×60d;对照1组(n=8):受者术后腹腔注射溶剂二甲基亚砜0.5ml×60d;对照2组(n=6):受者术后不给予任何处理。腹主动脉移植后60d取移植血管进行组织学观察,测量血管内膜厚度;免疫组织化学染色检测血管平滑肌细胞的增殖和迁移情况;酶联免疫(ELIA)法检测血清中血管内皮生长因子(VEGF)、γ干扰素(INF-γ)的变化。结果 验组与两个对照组比较,内膜增厚不明显且巨噬细胞浸润减少;血管平滑肌细胞增殖和迁移减少(P均〈0.01);两个对照组血清中VEGF、INF-γ表达显著高于实验组(P〈0.01)。结论 染料木黄酮在预防和治疗移植动脉硬化的过程中,对移植动脉血管平滑肌细胞的增殖和迁移有抑制作用,其机理可能与影响VEGF等的表达有关。 Objective By using rat orthotropic aortic allograft transplantation model, detect the preventive effects of genistein (Gen) on the proliferation and migration of the vascular smooth muscle cells and investigate potential mechanisms. Methods Aortic transplantation was performed with male Brown-Norway rats as donors and male Lewis rats as recipients. The recipients in the experiment were randomly divided into three groups: (1) experimental group (n = 8) receiving Gen at a dose of 20 mg/ (kg day) for 60 days continuously; (2) solvent control (n = 8) receiving 0. 5 ml analytical pure DMSO for 60 days; (3) control (n = 6) not subject to intervention. Histological changes were monitored 60 days after transplantation, and the average depth of intima was calculated by computer imaging analysis systems. Immunohistochemistry was used to detect the proliferation and migration of the vascular smooth muscle cells in the transplanted vessels,and ELISA to find the change of VEGF and INF-γ in different groups. Results Compared with two control groups, lymphocytes and macrophages infiltration,and the average depth of intima in experimental groups were markedly decreased (all P〈0. 001 ) and the proliferation and migration of the vascular smooth muscle cells in the transplanted vessels were less than the two control groups. The expression of VEGF in experimental group was lower than that in two control groups (8. 6731 ± 1. 0466 vs 21. 8340 ± 0. 7558 and 23. 2068 ± 1. 2028,P〈0. 01) as well as the INF-γ (23. 5282 ± 1. 9092 vs 162. 3535 ± 13. 1691 and 189. 5032 ±12. 7650, P〈0. 05). Conclusion During the inhibition of arteriosclerosis,Gen can prevent the proliferation and migration of the vascular smooth muscle cells in rat orthotropic aortic allograft transplantation model by influencing the expression of growth factor and cytokine.
