摘要
目的探讨Cks1在胃癌发生及其在Skp2调节p27Kip1降解过程中的作用。方法应用流式细胞术测定正常胃粘膜和胃癌组织中Cks1、p27Kip1、Skp2蛋白表达。结果胃癌组织中Cks1、Skp2表达明显高于正常胃粘膜(χ2=22.69,P<0.05;χ2=13.42,P<0.05),而p27Kip1表达则低于正常胃粘膜(χ2=14.83,P<0.05)。胃癌中Cks1、Skp2表达与p27Kip1表达呈负相关(r=-0.649,P<0.05;r=-0.732,P<0.05);而Cks1蛋白表达与Skp2蛋白缺乏相关性(P>0.05)。胃癌中Cks1的表达与肿瘤分化程度相关(χ2=5.05,P<0.05),而与肿瘤浸润深度、淋巴结转移及临床分期不相关(P>0.05)。结论Cks1可能参与了胃癌的发生;胃癌中Cks1可能参与了Skp2调节p27Kip1泛素化降解过程。
Objective To investigate the expression of Cksl protein and its relationship with the expression of Skp2, p27^Kip1 and tumor aggressiveness. Methods Flow cytometry was used to detect the expression of the three proteins in 20 cases of normal gastric tissue and 64 cases of primary gastric cancers. Results The expression of Cks1 and Skp2 in gastric cancers was higher than that in normal gastric tissue (x^2 = 22. 69, P〈0. 05; x^2 = 13. 42, P〈0. 05 respectively). While the expression of p27^Kip1 was lower than that in normal gastric tissue(x^2 = 14. 83,P〈0. 05). A significant inverse relation was observed between the expression of Cksl or Skp2 and of p27^Kip1 ( r= - 0. 649, P〈0. 05;r= - 0. 732, P〈0. 05, respectively). While there was no correlation between the expression of Cks1 and Skp2(P〈0. 05). The expression of Cksl was significantly associated with tumor differentiation. (x^2 = 5. 05, P〈0. 05). While it was not associated with depth of invasion, lymph node metasis and clinical stages (P〉0. 05). Conclusion Cks1may play an important role in human gastric tumorgenesis and it may be an essential accessory factor for Skp2 in protease-dependent destruction of p27^Kip1.
