内皮细胞一氧化氮合成酶多态性与血压、血脂和血糖的关系研究 被引量:1

摘要 目的研究内皮细胞一氧化氮合成酶(endothelial nitric oxide synthase gene,eNOS)298Glu/Asp多态性与血压、血脂和血糖代谢的关系。方法采用PCR-RFLP方法检测184例原发性高血压患者和196例血压正常对照者中eNOS298Glu/Asp多态性的分布,并分析在不同eNOS298Glu/Asp多态性下,血压与总胆固醇、TG、HDL、LDL、血糖等危险因素的相互关系。结果eNOS298Glu/Asp多态性在不同血压组和不同性别组中的分布没有显著差异。但不同年龄、舒张压、BMI组eNOS298Glu/Asp多态性分布有显著性差异(P<0.05);eNOS298突变型(Asp/Asp)和高血清胆固醇患者(血清总胆固醇5.4mmol/L以上)患高血压的相对危险度(OR)显著高于其他患者。结论eNOS298Glu/Asp多态性与血脂对舒张压有明显的联合作用,eNOS298突变型(Asp/Asp)能明显增加血清总胆固醇5.4mmol/L以上患者患高血压的危险性。eNOS298Glu/Asp多态性与血清总胆固醇水平结合,可用于预测和分析不同个体患高血压的危险性。 Objective To study the relationship of the polymorphism of endothelial nitric oxide synthase (eNOS) gene and blood pressure, lipid profiles and blood sugar level. Methods Totally 184 essential hypertension patients and 196 matched health individuals with normal blood pressure were subjected in this study. Their age, sex and BMI were recorded and eNOS Glu 298 Asp gene polymorphism were detected in using PCR-RFLP. Lipid profiles, including triglycerides ( TG), total cholesterol ( CHO ), HDL, LDL, and blood sugar level were also detected. Results Significant difference of the percentage of eNOS Glu 298 Asp gene were observed in age, systolic blood pressure, BMI group ( P 〈 0.05 ). Asp/Asp genotype significantly increased the risk of high systolic blood pressure in individuals with CHO above 5.4 mmol/L ( P =- 0.03, OR = 2.65 ). Conclusion eNOS Glu 298 Asp may modulate blood pressure by associating with lipid levels, eNOS 298 Asp/Asp increases the risk of hypertension in those with serum CHO over 5.4 mmoL/L, eNOS 298 Glu/Asp polymorphism and serum total CHO together may be used to predict and analyze the risk of having hypertension.
