摘要 目的通过等位基因酶的生化多态性可以发现小鼠DNA的序列多态性,为人类疾病基因鉴定和动物实验的研究提供丰富的动物基因库,为科研工作提供宝贵资源。方法应用国产生化标记检测试剂盒,采用遗传生化检测方法,对天津市卫生防病中心自主培育的近交系小鼠进行Akp-1(碱性磷酸酶-1)等二十五个生化位点标记基因检测。结果经检测发现TW(Tianjin wild)小鼠在Amy1(淀粉酶-1),Es2(酯酶-2),Es3(酯酶-3),Es10(酯酶-10),Gpd1(葡萄糖磷酸脱氢酶-1),Hbb(血红蛋白β链),Np1(核苷酸磷酸化酶-1),Pgm2(磷酸葡萄糖变位酶-2)共八个生化位点发现罕见基因。结论检测国内新品系的遗传生化基因位点,发现罕见基因型。 Objective TW mouse is a new domestic inbred strain, established by the Tianjin Center for Disease Control and Prevention. The aim of the present study was to carry out genetic characterization and detect rare genes in these inbred mice. Methods 25 alleles, including Akp-1 and others, were assayed in the inbred mice by genetic biochemical labeling technique using a homemade biochemical label detection Kit. Results Rare genes were detected at 8 biochemical loci: Amy1, Es2, Es3, Es10, Gpd1, Hbb, Np1 and Pgm2 in the TW mice. Conclusions Polymorphism of DNA sequences in the mice can be detected by biochemical polymorphism analysis with allele isomorphic enzymes. It may be of great importance to provide abundant animal gene bank to serve the identification of disease genes in humans and as an invaluable resource for experimental animal research as well.
