摘要
目的:观察加味桃核承气汤对四氯化碳(CCl4)肝纤维化的治疗作用,从分子生物学水平探讨其对金属蛋白酶组织抑制因子-1(TIMP-1)的影响。方法:用CCl4诱导大鼠肝纤维化,将实验动物随机分为正常对照组、模型对照组、加味桃核承气汤高、中、低剂量治疗组和秋水仙碱对照组;采用灌胃给药,疗程8周;用苦味酸-天狼红胶原染色方法观察肝纤维化积分(SSS)的变化;用免疫组化技术半定量检测肝组织TIMP-1的蛋白表达水平。结果:模型对照组SSS显著高于正常对照组,经加味桃核承气汤治疗后SSS显著减轻;模型对照组大鼠肝组织TIMP-1的蛋白表达水平增加。各治疗组的TIMP-1的蛋白表达水平均显著下降。结论:①加味桃核承气汤对CCl4诱导的肝纤维化有治疗作用。②加味桃核承气汤抗实验性肝纤维化作用机制与抑制肝星状细胞(HSC)的活化,调控TIMP-1的蛋白表达水平有关。
Objective: To study effects of Jiawei Taohechengqitang on expression of TIMP-1 protein in liver tissue of haptic fibrosis induced by CCl4 in rats. Methods: Hepatic fibrosis models were induced by CCl4 in rats. All the experimental rats were divided into six groups: normal control group, model control group, colchicine positive treatment group, high-dose Jaawei Taohechengqitang treatment group, meduim - dose Jiawei Taohechengqitang treatment group, low-dose Jiawei Taohechengqitang group. Drugs were fed for 8 weeks by intragastric administration. Scores of hepatic fibrosis were assessed with semiquantitive scoring system (SSS) through tianlanghong staining. Protein expression levels of TIMP-1 were determined with immunohistochemi- cal steining method. Results: SSS in model control group were higher than those in normal control group, but 10w erthou thosein -thre treatment groups with Jiawei Taohechengqitong. The protein expression levels TIMP-1 in the live tissues in model contrd group were markedly increased compared with thosein normal control proup, but its protein expression of TIMP-1 were markedly decreased in Jiawei Taohechengqitang groups. Conclusion: The one of mechanisms of Jiawei Taohechengqitang is lying in: It can reduce the expression levels of TIMP-1 protein.
