摘要
目的:观察加味桃核承气汤对四氯化碳(CCl4)肝纤维化的治疗作用,从分子生物学水平探讨其对金属蛋白酶组织抑制因子-1(TIMP-1)的影响。方法:用CCl4诱导大鼠肝纤维化,将实验动物随机分为正常对照组、模型对照组、加味桃核承气汤高、中、低剂量治疗组和秋水仙碱对照组;采用灌胃给药,疗程8周;用苦味酸-天狼红胶原染色方法观察肝纤维化积分(SSS)的变化;用免疫组化技术半定量检测肝组织TIMP-1的蛋白表达水平。结果:模型对照组SSS显著高于正常对照组,经加味桃核承气汤治疗后SSS显著减轻;模型对照组大鼠肝组织TIMP-1的蛋白表达水平增加。各治疗组的TIMP-1的蛋白表达水平均显著下降。结论:①加味桃核承气汤对CCl4诱导的肝纤维化有治疗作用。②加味桃核承气汤抗实验性肝纤维化作用机制与抑制肝星状细胞(HSC)的活化,调控TIMP-1的蛋白表达水平有关。
Objective： To study effects of Jiawei Taohechengqitang on expression of TIMP-1 protein in liver tissue of haptic fibrosis induced by CCl4 in rats. Methods： Hepatic fibrosis models were induced by CCl4 in rats. All the experimental rats were divided into six groups： normal control group, model control group, colchicine positive treatment group, high-dose Jaawei Taohechengqitang treatment group, meduim - dose Jiawei Taohechengqitang treatment group, low-dose Jiawei Taohechengqitang group. Drugs were fed for 8 weeks by intragastric administration. Scores of hepatic fibrosis were assessed with semiquantitive scoring system （SSS） through tianlanghong staining. Protein expression levels of TIMP-1 were determined with immunohistochemi- cal steining method. Results： SSS in model control group were higher than those in normal control group, but 10w erthou thosein -thre treatment groups with Jiawei Taohechengqitong. The protein expression levels TIMP-1 in the live tissues in model contrd group were markedly increased compared with thosein normal control proup, but its protein expression of TIMP-1 were markedly decreased in Jiawei Taohechengqitang groups. Conclusion： The one of mechanisms of Jiawei Taohechengqitang is lying in： It can reduce the expression levels of TIMP-1 protein.
