胰岛素受体基因NsiI多态性与原发性高血压的相关性研究 被引量:5

摘要 [目的]研究胰岛素受体(INSR)基因第8外显子NsiI多态性与原发性高血压病(EH)的关系。[方法]选择山东省五莲县15岁以上居民进行EH流行病学调查,检测相关临床指标,从中选取EH患者221例和正常对照204例,提取外周血白细胞DNA后进行PCR扩增,采用NsiI酶切和对部分PCR产物直接测序检测相关基因多态性。[结果]EH组与正常对照组除收缩压与舒张压有显著差别外,其他临床检验指标差异无统计学意义。EH组INSR基因N2等位基因频率高于正常对照组(P﹤0.05)。[结论]INSR基因NsiI多态性与中国人高压血病有关,N2等位基因可能是高血压病一个易感基因。 [Objective] To study the relationship between NsiI Polymorphism of insulin receptor gene (INSR) and essential hypertension (EH) . [ Methods] Epidemiological survey for essential hypertension was conducted in dwellers over 15 years old in Wu-Lian County, Shandong Province, and some correlated clinical markers were measured using. With random sampling, 221 EH patients and 204 normal controls were chosed to detect the NSiI pelymorphism of insulin receptor gene. DNA was extracted from peripheral blood leukocytes. Then pelymerase chain reaction (PCR) combined with restriction enzyme digestion was used to detect the pelymorphism. Finally the amplified gene was sequenced to certificate the mutant base. [ Results] The systolic bond pressure (SBP) and diastolic blood pressure (DBP) of hypertension group were significantly higher than those of control while other clinical features showed no difference between two groups; N2 allele frequency of INSR gene of EH group was higher than that of control (P〈 0.05) . [Conclusions] NsiI pelymorphism of INSR gene is associated with EH in Chinese population. N2 allele of INSR gene may be one of predisposing genes of hypertensive diseases.
