摘要
目的:探讨鳖甲煎丸对人肾小球系膜细胞(HMC)增殖及细胞外基质(ECM)不同成分的抑制作用。方法:应用细胞培养技术,进行人HMC培养,采取血清药理学方法,制备鳖甲煎丸药物血清,利用脂多糖(LPS)为刺激因子,应用四甲基偶氮唑蓝(MTT)法检测各组系膜细胞的增殖情况,并采用酶联免疫吸附试验(ELISA)法检测ECM中层黏连蛋白(LN)和Ⅳ型胶原(Col-Ⅳ)的含量。结果:LPS刺激后MC增殖和系膜基质中LN及Col-Ⅳ的分泌增多,与空白组比较有明显差异(P<0.01)。鳖甲煎丸能抑制正常条件及LPS刺激条件下MC增殖,并对ECM中LN、Col-Ⅳ的表达有抑制作用。结论:鳖甲煎丸能够抑制系膜细胞的增殖,抑制肾小球系膜基质中LN及Col-Ⅳ的表达,减少ECM的积聚,这可能是其防治肾小球疾病,延缓肾小球硬化的部分作用机制。
AIM: To investigate the role of Biejiajian Pills (BJJP) on proliferation of human mesangial cells (MC) and the expressive in extracellular matrix. METHODS : Cell culture technique and blood serum pharmacology method were used in experiment. MC were stimulated by lipopolysaccharide(LPS). The MC proliferation, secretion of laminin(LN) and collagen-Ⅳ(Col-Ⅳ) were measured by MTT, ELISA. RESULTS: LPS can promote MC proliferation, and secretion of LN and Col-Ⅳ. After stimulation of LPS, the secretion of LN and Col-Ⅳ increasd. MC proliferation was inhibited by BJJP pharmacological blood serum under normal and pathological condition. So as to the secretion of LN and Col-Ⅳ. CONCLUSION: BJJP can significantly inhibite the proliferation and secretion of LN and Col-Ⅳ, reduce the accumulation of ECM, which may be the mechanism of BJJP in treatment of glomcrulus disease and delay glomerular sclerosis.
