摘要 目的探讨水通道蛋白4(AQP-4)在大鼠创伤性脑水肿早期的表达及意义。方法按Feeney自由落体撞击法建立大鼠创伤性脑损伤(TB I)模型,用干/湿法测定大鼠TB I后不同时相脑组织含水量,并采用免疫组织化学方法检测脑组织AQP-4蛋白的表达.采用W estern B lot方法对AQP-4蛋白进行半定量分析。结果TB I后48小时伤侧脑水肿达高峰,同时水通道蛋白AQP-4在大脑半球表达降低(P<0.05),损伤侧大脑半球伤后48小时降低最为明显(P<0.01)。AQP-4的表达与脑组织水含量呈负相关。结论脑损伤后,AQP-4表达降低与脑水肿的形成密切相关。 Objective To investigate the dynamic change and the significance of cerebral aquaporin -4 (AQP-4) expression level during traumatic brain injury (TBI) in rats. Methods The experimental TBI model was established by bumpiness of free falling body according to Feeneys. The changes of brain water content (BWC) were measured by the methods of wet and dry weight. Immunohistohemistry and Western Blot were used to determine the change of expression of AQP -4 during different periods after TBI. Results Brain edema was maximal by 24h after TBI. Concurrently, AQP -4 protein expres- sion was decreased in hemispheres , being more pronounced in the traumatized hemisphere (250%) 48h after trauma. There was significantly negative correlation between protein expression of AQP -4. Conelusions Globally reduced AQP- 4 expression is closely related to the formation of cerebral edema during traumatic brain injury.
