摘要 目的对不同比例的壳聚糖-胶原(ch i-col)进行细胞相容性和细胞毒性的研究,以初步探讨其应用于牙周组织工程的可行性。方法将体外培养的人牙周膜细胞(PDLCs)接种于不同比例的ch i-col上复合培养,采用细胞计数评价PDLCs在ch i-col上的附着、生长情况,并通过MTT测试法和碱性磷酸酶(ALP)活性检测法研究ch i-col浸提液对PDLCs增殖和功能表达的影响。结果人PDLCs在ch i-col上形成良好贴附并增殖,而PDLCs在不同浓度的材料浸提液中的生长、增殖及ALP活性与对照组相比无统计学意义。结论ch i-col具有良好的三维空间结构和细胞相容性,且无细胞毒性,有望成为牙周组织工程的支架材料。 Objective The cytocompatibility and cytotoxicity of chitosan - collagen ( chi - col) were investigated to evaluate the feasibility of chi - col as the scaffold material of periodontal tissue engineerded on three - dimensional framework of chi - col. The cell growth in the scaffolds was observed by cell counting. And the influences of the chi - col extracts on cell proliferation and differentiation were observed by MTT assay and Alkaline phosphatase (ALP) activity assay. Results Cells adhered and proliferated on the three - dimensional framework of chi - col showed the porous structure of chi - col and the eugonic growth of cells in the scaffolds. The different concentrations of chi - col extracts had no influences on the cell proliferation and differentiation. Conclusions Provided with the porous structure and satisfactory cytocompatibility and without any cytotoxicity, chi - col is valued to further study as the scaffolds of periodontal tissue engineering .
