摘要
目的研究雷公藤甲素(Triptolide,Tri)对人晶状体上皮细胞(Hum an Lens Ep ithelial Cells,HLEC)增殖抑制作用。方法原代培养人晶状体上皮细胞,Tri不同浓度、不同作用时间后吖啶橙试剂(AO)染色,荧光显微镜下观察细胞核的形态变化;MTT法测定人晶状体上皮细胞增殖抑制率。结果雷公藤甲素对细胞的生长有明显的抑制作用,且呈浓度时间依赖性。结论一定剂量的Tri可抑制体外培养的人晶状体上皮细胞的增殖,为预防后囊混浊提供了新途径。
Objective To investigate the effect of Triptolide (Tri) on human lens epithelial cells (HLEC) proliferation in culture. Methods HLECs were cultured in vitro, the 3rd passage cells were seeded in 24 (96) well plates, and each well was put in 0. 1 ml of culture medium. The effects of different concentration of rift (40, 20, 10μg/L) on HLEC proliferation were evaluated with Fluorescence Microscope and MTT assaying method at 12h, 24h, 48h and 72h. Results Tri dosages ranging from 40, 20, 10μg/L could inhibit cellular proliferation effectively after incorporated 72h in a dose -dependent and time -dependent manner. 12h, 24h, 48h and effectively inhibition on HLEC proliferation. This could be beneficial to the prevention of posterior capsule opacification.
