罗格列酮对单侧输尿管梗阻大鼠肾间质纤维化的作用

摘要 目的探讨罗格列酮(RSG)对单侧输尿管梗阻大鼠(UUO)肾间质纤维化的作用。方法将24只大鼠随机分为假手术组、模型组、治疗组。对治疗组、模型组行单侧输尿管结扎术建立输尿管梗阻的动物模型。术后14天处死各组大鼠,取梗阻侧肾组织做HE和M asson染色及用免疫组化方法检测结缔组织生长因子(CT-GF)和α-平滑肌肌动蛋白(α-SMA)在肾组织的表达水平。结果罗格列酮可显著下调CTGF和α-SMA的表达,改善了肾脏病理改变。结论罗格列酮对UUO所致的肾间质纤维化的形成有抑制作用。 Objective To observe the roles of rosiglitazone in unilateral ureteral obstruction rats renal interstitial fibrosis. Methods Rats were randomly divided into rosiglitazone -treated group, UUO mod- el group and sham - operated group, respectively. The rats in the rosiglitazone - treated group and UUO model group underwent UUO. After 14 days of the operation, all rats were sacrificed and the resected kidneys were collected for HE and MASSON staining and immunohisto chemical staining for CTGF and α - SMA. Results In the obstructed kidneys, there was significant renal interstieial fibrosis and interstitial space expansion. Immunohistochemical staining showed that the expression of CTGF and α - SMA was significantly increased in the obstructed kidneys. In comparison with the model group , rosiglitazone could obviously decrease interstitial volume and the expression of CTGF and α - SMA. It also improved the histological changes of the UUO renal tissue. Conclusions Rosiglitazone can down - regulate the expression CTGF and.α -SMA in UUO rats. This demonstrated its potential ability to inhibit renal interstitial fibrosis caused by UUO in rats.
