摘要
用壳聚糖作载体,戊二醛为交联剂,采用吸附交联法将α-淀粉酶固定化,研究了固定化条件和固定化酶的性能.在酶浓度8g/L,6~8℃,pH6.8,吸附12h,l%戊二醛交联4h的固定化条件下,固定化效果较优.固定化酶的酸碱稳定性、热稳定性、贮存稳定性较好.最适作用pH较自由酶酸移,最适作用温度及Km值较自由酶高,同时显示出一定的操作稳定性.
?Amylase was immobilized on deacetylated chitin and crosslinked with glu taraldehydes.Enzyme concentration:8 mg/ml,temperature: 6~8°C,pH 6.8,absorbing time: 12 h,crosslinking time:4 h,glutaraldehydes concentration: 1 % was the optium condition for immobilization.The stability to pH,temperature,storage and the optium pH,the optium tem perature,the operational stability of immobilized α- amylase were studied.
