胰岛素抵抗糖尿病KKAy小鼠肌肉和肝组织PTEN蛋白表达增强 被引量:6

摘要 目的观察胰岛素抵抗KKAy小鼠胰岛素敏感组织肌肉及肝组织PTEN蛋白的表达,了解PTEN在胰岛素抵抗机制中的作用。方法实验分为对照组1(C57BL普通饲料喂养组),对照组2(C57BL高脂饲料喂养组),KKAy小鼠组。均尾静脉血测末梢血糖,连续两周随机血糖>16.7 mmol/L诊断为糖尿病,取等量股四头肌、肝组织进行Western blot检测。结果与对照组相比,KKAy组股四头肌、肝PTEN蛋白均明显增多(P<0.01)。结论PTEN可能是一种重要的负性调节因子,调节三磷酸肌醇激酶(PI3Ks)/蛋白激酶B(Akt)所介导的信号传导通路,在胰岛素抵抗的发生、发展中起着重要的作用。 Objective To detect protein PTEN in live and muscle tissue of KKAy diabetic mice and to investigate whether PTEN is associated with the insulin resistance in diabetic KKAy mice. Methods Animals were divided into normal diet C57BL group (n =7 ,sixteen-week-old C57BL mice), high fat diet C57BL group (n =7), diabetic KKAy group (n =7), the latter two groups were fed with normal diet until week 12, followed by high fat diet for 4 weeks. Blood glucose was measured every week, it would be diagnosed of diabetes if blood glucose more than 300 mg/dl (16. 7 mmol/L) in two consecutive weeks. Dispatched the mice ,took quadrieeps of femoris muscle and liver tissue, added tissue lytie solution, measured the protein concentration by Bradford method and detect the PTEN protein expression in muscle and liver tissue by Western bolts methods. Results The PTEN protein level was signi.fieantly increased in quadrieeps muscle and live tissue in KKAy mice as compared to the both age-matched C57BL control groups(P 〈 0. 01 ). Condusion PTEN might be a negative modulator of the PI3K-Akt pathway and then plays an important role in the development of insulin resistance.
