胰岛素样生长因子及其I型受体抗体对人肾上腺皮质癌SW-13细胞株体外生长的影响 被引量:2

Effect of insulin-like growth factor and its receptor-I antibody on growth of human adrenocortical carcinoma SW-13 cell lines in vitro

摘要 目的探讨胰岛素样生长因子及其I型受体抗体对人肾上腺皮质癌SW-13细胞株体外生长的影响。方法采用MTT法描绘不同浓度胰岛素样生长因子及其I型受体抗体作用后SW-13细胞株体外培养生长的促进及抑制曲线,并比较不同浓度胰岛素样生长因子和其I型受体抗体组及单一胰岛素样生长因子或其I型受体抗体组与阴性对照组之间的差异。结果胰岛素样生长因子能明显促进SW-13细胞增殖,其作用效果与作用浓度在一定浓度范围内呈正相关(P<0.05);而胰岛素样生长因子I型受体抗体能抑制胰岛素样生长因子对SW-13细胞的刺激作用,并且对SW-13细胞的增殖亦有直接抑制的作用(P<0.05)。结论胰岛素样生长因子可促进SW-13细胞株的生长,而这种作用亦是通过胰岛素样生长因子I型受体传导;胰岛素样生长因子I型受体抗体可明显抑制这种生物学作用,可能在肾上腺皮质癌的治疗中发挥重要作用。 Objective To evaluate the effect of insulin-like growth factor (IGF) and its receptor-Ⅰ antibody on the growth of human adrenocortical carcinoma SW-13 cell line in vitro. Method The growth curves of SW-13 cell treated with IGF and its receptor-Ⅰ antibody were obtained by means of MTT assay. The effects of the two agents, added either alone or in combination at different concentrations, on the cell growth were evaluated. Results IGF significantly promoted proliferation of SW-Ⅰ3 cells, and its effect was positively correlated with its concentrations (P〈0.05). IGF receptor-Ⅰantibody inhibited the effect of insulin-like growth factor with direct inhibitory effect on proliferation of SW-13 cells (P〈0.05). Conclusion IGF can promote the growth of human adrenocortical carcinoma SW-13 cells via its receptor-Ⅰ. IGF receptor-Ⅰ antibody can inhibit the effect of the growth factor, suggesting a possible role of this receptor in the treatment of adrenocortical carcinoma.
