摘要
目的检测大鼠局灶性脑缺血再灌注不同时相脑皮质及皮质下多胺含量的变化,动态观察局灶性脑缺血再灌注后多胺含量变化的时相规律。方法在Longa线栓法的基础上进行改良,复制大鼠2 h局灶性脑缺血再灌注2、4、8、24 h动物模型,用高效液相色谱法测定脑缺血区皮质和皮质下不同时相多胺的含量,分析其时相变化规律及意义。结果实验组大鼠脑缺血区皮质和皮质下腐胺含量于再灌注4 h后即开始逐渐升高(P<0.05),高峰出现在再灌注2 4 h后(P<0.05);再灌注8、2 4 h实验组精脒、精胺含量较对照组明显下降(P<0.05)。结论局灶性脑缺血再灌注后腐胺含量增高,精脒、精胺含量减少,共同参与了脑缺血损伤。
Objective To study the changes of polyamine levels in the cortex and subcortex at different times of reperfusion after 2 hours focal cerebral ischernia in rats in order to explore the regularity and significance of these changes. Methods The animal model of 2 hours reversible focal cerebral ischemia was estalblished in rats by ameliorating the method of Longa's intraluminal suture occlusion of middle cerebral artery (MCA). The changes of polyamine levels in the cortex and subcortex were measured by high-performance liquid chromatograph (HPLC) at 2, 4, 8 and 24 h after reperfusion respectively. The data and their significances were analyzed. Results Compared with the control group, putrescine level of the experimental group increased at 4h after reperfusion ( P〈0.05), the peak value appeared at 24 h after reperfusion (P〈0.05) ; sperrnidine and spermine levels decreased at 8 h and 24 h after reperfusion in the experimental group ( P 〈0.05). Conclusion After reperfusion following transient focal cerebral ischemia, the putrescine level increases while the spermidine and spermine levels decrease . All these changes may participate in the brain damage after cerebral ischemia.
