预处理对乳鼠心肌细胞缺氧复氧损伤的影响及其机理探讨

摘要 目的观察预处理(PC)对乳鼠心肌细胞缺氧复氧(A/R)损伤的影响,并探讨其机理。方法采用心肌细胞A/R损伤模型,用短暂缺氧进行预处理。结果预处理能提高A/R后心肌细胞存活率,减少细胞MDA产生及LDH和蛋白质的漏出,激活心肌细胞PKC活性。PKC抑制剂完全阻断了PC的细胞保护作用。结论离体心肌细胞存在PC保护现象。 Objective:To investigate the effect of hypoxic preconditioning(HPC) on anoxia-reoxygenation(A-R) injury of neonatal rat cardiomyocyte A-R,HPC was performed by transient hypoxia.Results:HPC inproved viability of cardiomyocyte during A-R( P <0.01),reduced formation of MDA and leakage of LDH and protein in cells.HPC also activated cardiomyocyte PKC.H (an inhibitor of PKC) abolished the above protection of HPC.Conclusion:There exists protective phenomenon of preconditioning in cardiomyocyte of neonatal rat in vitro and the mechanism involves in activation of PKC.
