粒细胞集落刺激因子影响白血病细胞增生的研究 被引量:1

摘要 目的探讨粒细胞集落刺激因子(G-CSF)对急性白血病(AL)细胞增生的影响。方法急性髓系白血病(AML)患者30例,急性淋巴细胞白血病(ALL)患者20例,正常对照20例。化疗前留取骨髓5ml,制备其单个核细胞(MNC)悬液,并分别加入不同浓度G-CSF培养24h后用流式细胞仪(FCM)检测其DNA倍体的量。同时检测MNC的G-CSF受体(G-CSFR)表达率。结果骨髓MNC培养24h后各浓度的DNA倍体量在AML随着G-CSF浓度的增加有线性上升趋势,在ALL和正常对照呈直线。骨髓MNCG-CSFR表达率AML(68.59±13.99)%;ALL(1.90±0.93)%;正常对照(70.50±10.80)%。结论G-CSF可促进AML细胞增生,对ALL和正常粒细胞增生无促进作用。G-CSFR于恶性及正常粒细胞表达,淋巴细胞不表达。 Objective To study of mononuclear proliferation of AL under the G-CSF stimulation. Methods AML 30 cases, ALL 20 cases, normal control 20 cases were chosen. Aspiration bone marrow 5 ml before chemotherapy to make out MNC liquid to cultivate 24 hours to join different concentration G-CSF inspectively, and to survey their DNA diploid with FCM, Meanwhile to measure the expression of G-CSFR in MNC with FCM. Results The different G-CSF concentration in AML is linearity increasing with DNA diploid, in ALL and normal control is linearity horizon with DNA diploid. The expression rate of G-CSFR: AML is (68.59±13.99)%; ALL is (1.90±0.93)%; normal control is(70.50±10.80)%, respectively. Conclusion G-CSF can promote proliferation of AML cells and can not promote ALL and normal granuloeyte preliferation. G-CSFR express in AML and normal granulocyte and do not express in ALL.
