摘要
为了解空间诱变产生的水稻核雄性不育突变体ws-3-1基因表达的特点,对ws-3-1和原种特籼占13减数分裂二分体时期的cDNA进行了抑制缩减杂交(SSH)分析,分别从正向和反向缩减文库中检测到109和104个表达有差异的克隆.对这些克隆进行了测序和同源性分析,并利用Northern杂交技术,从中鉴定出5个在原种与突变体间减数分裂二分体时期表达有差异的基因,并分析了该雄性不育突变体基因表达的变化与表型的关系.
Differentially expressed genes between rice male sterility mutant ws-3-1, induced by space flight, and Texianzhan 13, it's original indica rice variety, at the meiotic stage were analyzed by cDNA suppression subtractive hybridization (SSH). Forward- and reverse- subtractive libraries were constructed by using cDNAs from Texianzhan13 or ws-3- 1 as tester. By using library screening with Southern blot-ting, 109 and 104 positive clones with different transcriptional abundance between Texianzhan13 and ws- 3-1 were selected from the forward- or reverse-subtractive libraries respectively. Northern blotting analysis was performed to compare the transcription level between ws-3-1 and Texianzhan13, and difference in ex-pression abundance was found in five cDNA clones. The differences in the expression of these genes and their relationship with male sterility were discussed.
