特应性皮炎患儿淋巴细胞Th1/Th2相关趋化因子受体的表达

Detection of chemokine receptors related with Th1/Th2 on peripheral blood lymphocytes of patients with atopic dermatitis

摘要 目的 在单细胞水平上,探讨几种与Th1/Th2相关的趋化因子受体的表达在特应性皮炎(AD)发病机理中的作用。方法 采用荧光双色流式细胞术分析35例AD患儿及34例健康对照外周血CD4^+及CD8^+淋巴细胞表面与Th1细胞相关的趋化因子受体CXCR3、CCR5和Th2细胞相关的趋化因子受体CCR3、CCR4及CCR8的表达。结果 与健康对照组相比,AD患儿外周血淋巴细胞表面CXCR3(87.10%±10.00%)、CCR5(11.20%±5.16%)、CCR3(12.00%±7.46%)和CCR4(25.90%±8.19%)表达水平显著增加(P分别〈0.001、〈0.05、〈0.001、〈0.05),进一步分析发现,CD4^+淋巴细胞表面CXCR3(24.30%±7.60%)、CCR5(2.17%±1.17%)、CCR3(4.06%±2.34%)和CCR4(16.80%±4.10%)表达水平明显升高(P〈0.05),CD8^+淋巴细胞表面CXCR3(22.60%±6.45%)、CCR5(3.28%±1.80%)、CCR3(4.26%±2.02%)、CCR4(15.50%± 4.87%)和CCR8(1.30%±0.59%)表达水平亦明显升高(P〈0.001或P〈0.05)。结论 不同淋巴细胞表面Th1/Th2相关的趋化因子受体表达的增加,可能导致Th1/Th2免疫反应在高水平状态下发生失衡,从而在AD发病机理中起着重要作用。 Objective To study the chemokine receptors related with Th1/Th2 in the patients with atopic dermatitis(AD). Methods The chemokine receptors CXCR3, CCR5, CCR3, CCR4 and CCR8 on peripheral blood lymphoeytes of 35 patients with AD and 34 normal controls were analyzed by flow cytometry with a two color immunofluorescent staining. Results CXCR3 (87. 10% ± 10.00%), CCRS(11. 20%±5. 16%), CCR3(12.00%±7.46%) and CCR4(25.90%±8. 19%) on lymphocytes were significantly higher in the patients with AD than in the controls(P〈0. 001 or P〈 0.05). There were significant difference in CXCR3(24. 30% ±7.60%), CCR5(2. 17% ± 1. 17%), CCR3 (4.06 ± 2.34 % ) and CCR4 ( 16.80 % ± 4. 10 %) on CD4^ + Tlymphocytes between the patients and the controls (P〈0.05) ,meanwhile, the number of CXCR3(22.60%±6.45%), CCR5(3.28%± 1.80%) ,CCR3(4.26%±2.02%),CCR4(15.50%±4.87%) and CCR8 (1.30%±0. 59%)-positive CD8^+ -lymphocytes in peripheral blood was increased in the patients with AD compared to the controls (P〈0. 001 or P〈0.05). Conclusion The expressions of CXCR3, CCR5, CCR3, CCR4 and CCR8 on different lymphocytes increase in peripheral blood of AD patients. It may break the balance of Th1 and Th2 immune response and play an important role in the immune response to AD.
