摘要
目的了解老年期缺血肾分子调控水平的变化。方法应用Northernblot和Slotblot分子生物学杂交技术,在老年大鼠急性缺血再灌注1、4、8小时肾组织中,研究了即刻早期反应基因之一c-fos的表达,并与青年鼠作了比较。结果老年及青年鼠肾组织中c-fos信使核糖核酸(mRNA)表达高峰均在缺血再灌注1小时,但两者的增高幅度有明显差别,老年鼠增高仅10.2倍,青年鼠增高近20倍,再灌注4和8小时青年、老年鼠c-fosmRNA表达均接近正常。结论 在分子调控水平反应了老年期缺血肾修复潜能的低下,这可能是老年人缺血性急性肾功能不全恢复不良的部分原因。
Objective To identify age-related changes of regulatory mechanism associated with the molecular events following renal ischemia. Methods Northern blot and slot blot analysis were used to measure the levels of growth-related immediate early gene-c-fos mRNA in the ischemic-reperfusion rat kidney. Young and aged rats underwent 45 minutes of renal artery occulusion followed by 1- , 4-and 8-hour, reperfusion and the expression of c-fos mRNA was examined. Results (1) Levels of c-fos mRNA in both the young and aged kidneys increased obviously after one hour of ischemia; (2)The ratio of c-fos mRNA level of 1-hour reperfused kidney to c-fos mRNA level of sham-operated kidney was lower in aged rats as compared with young ones ; (3) There was no apparent change in the expression of c-fos mRNA at 4-or 8-hour of reperfusion. Conclusions The results suggest that the potentiality of renal recovery from acute ischemia damage was lower in aged kidney.
