Determination of metallothioneins by resonance light scattering method using alizarin red

摘要 目的:建立一种灵敏、简便的测定金属硫蛋白(MTs)的共振光散射(RLS)新方法。方法:在pH2.5Britton—Robinson缓冲溶液中,在十二烷基苯磺酸钠(SDBS)存在下,茜素红(AR)与MTs作用,导致体系的RLS明显增强。结果:在实验最适条件下,MTs—AR—SDBS体系的共振光散射强度随着MTs的加入在505nm处急剧增强,且增加的强度与MTs加入量成线性关系(r=0.9994)。方法的线性范围为0.06~16.5μg/ml,检出限为19.05ng/ml。结论:方法简便、灵敏、选择性好,检测加标样品中MTs,结果较满意。 Objective:To develop a sensitive and simple resonance light scattering method for determination of metallothioneins (MTs). Methods: In britton - robinson buffer solution of pH 2. 5 and the presence of sodium dodecylbenzene sulfonate (SDBS) , the intensity of system, sresonance light scattering (RLS) was obviously enhanced because of interaction of MTs with alizarin red (AR). Results:Under the optimal experimental conditions,the enhancement of resonance light scattering of the MTs - AR - SDBS system at 508 nm was proportional to the concentration of MTs in the range of 0. 06 - 16. 5 μg/ml ( r = 0. 9994 ). The detection limit was 19. 05 ng/ml. Conclusion :The proposed assay succeeds in spiked sample determination with the advantages of simplicity and high sensitivity and good selectivity.
