摘要
目的:研究海(水)产品中霍乱弧菌新的增菌分离检验方法,提高方法的最小检出量及结果的准确性和可靠性。方法:海(水)产品等水生动物检样经搅碎后,应用四硼酸钠缓冲溶液,将污染在组织中的霍乱弧菌等微生物洗脱溶释游离在液体内,吸取上层清液加入5倍浓碱性蛋白胨,霍乱弧菌在特定的条件下进行有效增菌,然后分离鉴定。结果:四硼酸钠能有效地控制增菌条件。pH值变异系数0.63%,减少干扰物质组胺75.0%-80.0%,最小检出量0.01-0.04cfu/g,比原方法可提高5个对数的阳性检出量,检出率提高50%以上。结论:本方法具有防腐、抗干扰强、增菌条件稳定、分离平板上杂菌少、菌落易识别、菌型不易变异、操作方便和阳性检出率高的特点,可缩短检测周期1倍,节约成本65.0%~70.0%。不但适用于海(水)产品,也适用于其它固体食品的检测。
Objective:To improve the sensitivity, accuracy and reliability of testing methods of Vibrio cholera in marine products. Methods:The samples of marine product were smashed and washed with borax buffer. The upper solution was drawn, and basic peptone was put in. The germ was amplified, separated and identified. Results:Borax was effective to control germ amplification. The coefficient of pH variation was 0. 63%, histamine was reduced by 75.0% - 80. 0%, the sensitivity was 0.01 - 0. 04 cfu/g, the rate that Vibro cholera was properly test increased by more than 50%. Conclusion :This method is characterized by antiseptic, antiinterference, steadiness, convenience and high rate that Vibrio cholera was properly test. The testing period is reduced by half, the cost is reduced by 65. 0% - 70. 0% , and this method is also suitable for testing other solid food.
