乳腺导管增生癌变过程中HIF-1α的表达变化及意义 被引量:8

摘要 目的探讨乳腺导管增生癌变过程中缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)的表达变化及其意义。方法应用免疫组织化学方法检测HIF-1α在乳腺正常组织及各组病变中的蛋白表达情况。结果乳腺导管原位癌和浸润性导管癌组织中HIF-1α蛋白阳性表达率明显高于其他组织(P<0.01);浸润性导管癌组织中HIF-1α蛋白阳性表达率明显高于乳腺导管原位癌,且其表达与淋巴结转移、组织学分级、TNM分期密切相关。结论HIF-1α的表达强度在一定程度上与细胞的恶性倾向有关,HIF-1α参与了乳腺导管增生癌变的演进过程。 Objective To investigate the expression level of HIF-1α during breast carcinogenesis and tumor progression and its clinicopathological significance. Methods Expression of HIF-1α in invasive breast carcinomas ( n = 74 ), adjacent ductal carcinoma in situ(DCIS) ( n = 13 ), DCIS alone( n = 5 ), ductal atypical hyperplastic lesions( n = 24 ), usual ductal hyperplastic lesions (n = 35 )and normal breast epithelia ( n = 46) was analyzed by immunohistochemistry using specific monoclonal antibodies. Results HIF-1α was overexpressed in tumor specimens as compared with normal epithelia and ductal hyperplastic lesions. The expression of HIF-1α in invasive breast carcinomas was significantly higher than that in DCIS. HIF-1α expression level was correlated with TNM stage, lymph node metastasis and histological grade( P 〈 0.05 ). Conclusion HIF-1α may play an important role in human breast cancer progression.
