摘要
用MA—104细胞,以静止培养条件,从1月龄内的犊牛腹泻粪便中,分离出4电轮状病毒。经传至4~6代均使感染细胞出现特征性细胞病变。经直接电镜和免疫株镜观察,见有典型的轮状病毒颗粒,其RNA电泳型与NCDV株一致,以抗NCDV株高免血清琼扩检测及轮状病毒ELISA检测均为阳性。这4株病毒分别命名为DNRV_1,DNRV_2,DNRV_3,DNRV_4。
Four rotavirus strains (designated DNRV-1, DNRV-2, DNRV-3, DNRV-4) were successfully isolated from the feces of about one-month-old calves with diarrhea. MA-104 cell and stationary-tube culture were used for the replication and isolation of the virus. After three or four generations in MA-104 cell, all the isolated strains could produce obvious cytopathic effects(CPE). The direct electron microscope and immune electron microscope revealed the presence of particles with typical rotavirus morphology; The isolated strains were identical with NCDV in RNA electrophoresis. By the methods of AGP and ELISA assay with anti-serum to NCDV, the results were positive in all the isolated strains.
