膈下逐瘀汤对肝癌Bel-7402细胞增殖的抑制及与磷酸化AKT的相关性研究 被引量:4

摘要 目的:观察膈下逐瘀汤对肝癌Bel-7402细胞增殖的抑制及与磷酸化AKT的相关性研究。方法:膈下逐瘀汤配成浓度分别为:膈下逐瘀汤(C1):0.25×10^(-4)g/mL,膈下逐瘀汤(G2):0.5×10^(-4)g/mL,膈下逐瘀汤(G3):1.0×10^(-4)g/mL,膈下逐瘀汤(G4):2.0×10^(-4)g/mL;顺铂配制成浓度分别为:顺铂(S1):0.5μg/mL,顺铂(S2):1.0μg/mL,顺铂(S3):1.5μg/mL,顺铂(S4):2.0μg/mL,作用于Bel-7402细胞,用噻唑兰(MTT)比色法检测膈下逐瘀汤各组对Bel-7402细胞生长的影响,并且与顺铂各组相对照,用蛋白印迹试验(Western blot- ting analysis)检测不同浓度药物作用48h、72h时磷酸化AKT的表达水平。结果:膈下逐瘀汤对肝癌Bel-7402细胞的增殖及磷酸化AKT的表达均有显著的抑制作用,膈下逐瘀汤浓度在2.0×10^(-4)g/mL时其48h细胞生长抑制率介于S1组与S2组之间。结论:膈下逐瘀汤抑制肝癌Bel-7402细胞增殖的机制可能是通过抑制了细胞内磷酸化AKT这一途径而起作用。 Objective:To observe the effect of Gexia Zhuyu Tang(GZT,膈下逐瘀汤) for the inhibition of liver cancer Bel-7402 een proliferation and phosphorylated AKT. Mothods:GZT was prepared as different concentrations:0.25×10^(-1)g/ml,0.5×10^(-4)g/ mL,1.0×10^(-4)g/mL and 2.0×10^(-4)g/ml,Cisplatin was prepared as different concentra- tions:0.5μg/mL(SI),1.0μg/mL(S2),1.5μg/mL(S3)and 2.0μg/mL(S4)acting on Bel-7402 cells.The influence of various concentrations of GZT on the growth of Bel - 7402 cells were determined by MTT colorimetric method and compared with various cisplatin groups,the expression level of phosphorylated AKT in 48h and 72h of different concentra- tions were determined by western blotting analysis,Results:GZT can markedly inhibit Bel-7402 cells proliferation of liver cancer and expression of phosphorylated AKT,the concentration of GZT in 2.0×10^(-4)g/ml.the 48h inhibition rate on cells growth was between S1 and S2.Conclusion:The mechanism of GZT inhibition on Bel-7402 cells proliferation may be related to intracellular phosphorylated AKT.
