蒺藜总皂苷对家兔主动脉内膜粥样硬化病变的影响

Effects of GSTT on early atherosclerotic lesion of aortic intima in rabbits

目的:观察蒺藜总皂苷对高脂血症家兔的血脂及主动脉内膜粥样硬化病变的影响,并探讨其对动脉粥样硬化的治疗作用。方法:取50只健康雄性日本大耳白兔,随机取10只作为正常对照组,喂普通饲料,其余40只给予高脂饮食喂养4周后,再随机分为4组:①模型组10只,继续喂高脂饲料;②小剂量治疗组10只,喂以高脂饲料和蒺藜总皂苷6.3m g/kg.d;③大剂量治疗组10只,喂高脂饲料和蒺藜总皂苷12.6m g/kg.d;④阳性药物对照组,喂以高脂饲料和辛伐他汀2.35m g/kg.d,2周后,测定血脂;光镜和电镜下观察主动脉内膜形态变化。结果:形态学观察发现:模型组主动脉内膜出现典型粥样硬化病变,给药组主动脉内膜受损程度明显减轻。电镜显示:模型组中膜平滑肌细胞增殖,转化为合成表型,有新生内膜形成。而治疗组明显减轻。结论:蒺藜总皂苷具有保护主动脉内膜,干预动脉粥样硬化病变发生的作用。

Objective：To investigate the effects of gross saponins from Tribulus terrestris （GSTT）on the hyperlipemia and atherosclerotic lesion of aortic intima in rabbits. Methods ：Fifty healthy male Japanese white rabbits were used in the experiment, 10 of which were randomly selected and allocated as normal control group,fed with normal diet. Four weeks after the other forty rabbits fed with hypercholesterol diet for, they were randomly divided into four groups ：the model group, low-dose GSTT group, high-dose GSTT group and positive control group, and treated with hypercholesterol diet,GSTT 6. 3mg/kg·d, 12. 6mg/kg·d and simvastatin 2.35mg/kg·d in,respectively. Two weeks after administration,the blood liqid of rats in above groups was detected and the morphological changes of the aortic intima were observed under light and electron microscopes. Results： The morphological observation showed that typical atherosclerotic lesions of aortic intima appeared in the model group, while the degree of atherosclerotic lesions was obvious lighter in the group treated with GSTT compared with the model group. Electron microscropy showed that the early neotimal lesions were characterized by intimal thickening and the presence of SMCs in the model group compared with the therapy group. Conclusion：GSTT can protect the aortic intima and interfere the occurrence of atherosclerotic lesion.
