摘要
目的探讨应用TGF-β1多克隆抗体抑制术后屈指肌腱黏连的效果。方法将108只来亨鸡随机分成3组,行中趾屈指肌腱切断吻合术,术中分别滴加生理盐水或不同浓度的TGF-β1多克隆抗体,于术后1、3、8周取肌腱行HE染色、Masson胶原染色、苦味酸-天狼星红染色和TGF-β1免疫组化染色。结果TGF-β1多克隆抗体10μg/mL组炎症过程缩短,塑形过程加速;TGF-β1多克隆抗体5μg/mL组、10μg/mL组Ⅰ胶原含量减少,且Ⅰ/Ⅲ胶原比例下降,TGF-β1的表达降低。结论TGF-β1多克隆抗体可以有效地抑制TGF-β1在肌腱损伤修复过程中的作用,减少肌腱的黏连、瘢痕组织的形成,这可能是治疗屈指肌腱损伤术后黏连潜在的有效方法之一。
Objective To investigate the preventive effect of TGF-β1 neutralizing antibody on flexor tendon adhesion from operation. Methods Totally 108 leghon cocks performed anastomosis operation were divided into three groups randomly: normal saline (control group), 5μg/mL group, and 10μg/mL group of TGF-β1 antibody. At week 1, 3, and 8, respectively, after operation, HE staining, Masson staining, Sirius red-polarization staining and TGF-β1 immunohistochemistry staining were used. Results Compared with the control group, the 10μg/mL group had less shortened progress of inflammation and accelerated progress of molding; in the experimental groups (5μg/mL group, 10 μg/mL), the density of the collagen Ⅰ type was smaller, the ratio of Ⅰ/Ⅲ collagen and expression of TGF-β1 decreased. Conclusion TGF-β] multiclonal neutralizing antibody can effectively inhibit the function of TGF-β1 during the flexor tendon repair, reduce tendon adhesion and scar formation, suggesting that it is a latent and effective method for preventing flexor tendon from adhering after operation.
