黄芪对肺纤维化大鼠MMP-2和TIMP-1表达的影响 被引量:39

摘要 目的观察黄芪干预肺纤维化形成的作用,初步探讨其作用机制。方法SD大鼠36只,随机分为3组,模型组和干预组气管内注射博莱霉素诱导肺纤维化,空白组在相同条件下给予生理盐水。第2天起干预组均腹腔给予黄芪注射液,其余各组在相同条件下给予生理盐水。治疗第7天和第28天,用RT-PCR和免疫组化ABC法观察各组鼠肺组织中基质金属蛋白酶-2(MMP-2)和金属蛋白酶组织抑制剂-1(TIMP-1)表达的变化。结果干预组第7天和第28天肺组织中MMP-2和TIMP-1 mRNA的表达量均较模型组减少(均P<0.01),免疫组织化学检测也显示出同样的结果(P<0.05)。结论黄芪干预肺纤维化形成的机制可能是其能在一定程度上调整MMP-2和TIMP-1比值使其趋于平衡,延缓甚至抑制纤维化的进程。 Objective To investigate the intervention of Radix astragali to pulmonary fibrosis and the mechanism. Methods Thirty-six Sprague-Dawley rats were randomly divided into three groups. The model group and the intervention group were injected with bleomycln endotracheally to induce pulmonary fibrosis, while the empty group with normal saline instead in the same condition. From the second day the intervention group was injected with Radix astragali intraperitoneally, while the other two groups with normal saline instead. On the 7th day and 28th day the lungs were isolated. The expression of MMP-2 and TIMP-1 mRNA and protein was detected by RT-PCR and immunohistochemical method respectively. Results The expression of MMP-2 and TIMP-1 mRNA and protein in Radixastragali intervention group was decreased on the 7th and 28th day as compared with that in the model group. Conclusion The intervention mechanism of Radix astragali to pulmonary fibrosis is probably contributed to the regulation of MMP-2 and TIMP-1 balance to some extent, and alleviation, even suppression of pulmonary fibrosis process.
