高效液相色谱—蒸发光散射测定杞黄颗粒中黄芪甲苷的含量

Determination of Astragaloside Ⅳ in Qihuang Granules by HPLC-ELSD

目的制定杞黄颗粒中黄芪甲苷的含量测定方法。方法用HPLC—ELSD法，Shim—pack C18-ODS色谱柱，流动相乙腈-水（32：68），流速1．0ml／min；ELSD参数：漂移管温度40℃，气流压强：4．1Bar。结果回归方程为Y=1．614X＋4．2577，r=0．9996，线性范围为2．036—7．635μg。回收率为97．55％，RSD为2．02％。结论本研究建立的方法简便、可靠、准确，可以作为杞黄颗粒的质量控制方法。

Objective To determine astragaloside Ⅳ in Qihuang Granule by HPLC-ELSD. Methods HPLC-ELSD was used in the quantitative analysis by using Shim- pack C18-ODS chromatography column and acetonitril- water（32： 68） as a mobile phase. The flow rate of mobile phase was 1.0 ml/min. The drift tube temperature was 40℃. The gas pressure was 4. 1Bar. Results The regression equation was Y = 1. 614X ＋ 4. 2577, r = 0. 999 6 （2. 036 - 7.635 μg）. The average recovery of astragaloside Ⅳ was 97.55% ,RSD was 2.02%. Conclusion The method is simple ,reliable ,accurate and can be used as the quality control method of Qihuang Granule.
