缝隙连接蛋白介导的细胞间通讯与胶质瘤自杀基因治疗

摘要
1967年Kevel首次在大鼠心肌细胞问和肝细胞问发现了一种构造精妙的连接方式,并将之命名为“缝隙连接”(gap junction,GJ)。中枢神经系统细胞问存在有多种特异性连接蛋白(connexin,CX)的分布和表达并与中枢神经系统多种疾病的发生、发展密切相关,如胶质细胞瘤、脑膜瘤、癫痫、脑缺血缺氧性损伤及Alzheimer病等。CX、GJ及由此构成的细胞内信号转导(gap junction intercellular communication,GJIC)通路在肿瘤自杀基因治疗“旁观者效应”(bystandereffect,BE)中发挥着极为重要的作用。GJ及GJIC的生理病理变化直接影响脑胶质瘤细胞的增殖、浸润及凋亡。
