甲基汞对肺癌细胞的增殖抑制作用

摘要 目的探讨甲基汞对小鼠Lewis肺癌细胞的增殖抑制作用。方法采用MTT比色法。对照组仅加培养液;顺铂组、As2O3组、甲基汞组分别加入不同浓度的顺铂、As2O3、甲基汞,用DG-3022A型酶联免疫检测仪测定光密度(A)值,计算增殖抑制率,并以倒置光学显微镜观察各组Lewis肺癌细胞生长密度及形态变化。结果甲基汞对小鼠Lewis肺癌细胞有较强的增殖抑制作用,并存在着时间和剂量效应关系,而且同一时间各浓度之间、同一浓度各时间之间均存在显著性差异(P<0·001);甲基汞的增殖抑制作用强于顺铂和As2O3,起效浓度较后两者低,发挥作用较后两者快。结论甲基汞对小鼠Lewis肺癌细胞有增殖抑制作用,并存在着时间和剂量效应关系,其增殖抑制作用强于顺铂和As2O3。 Objective To investigate the inhibition of methyl mercury on the proliferation of rats Lewis lung cancer cells and to compare with As203 and cisplatin. Methods MTT chromatometry was adopted. Cultured Lewis cells were stimulated with different doses of methyl mercury, As2O3, cisplatin at 40, 20, 10, 5, 2. 5 μmol/L. The optical density (A) value was detected by DG-3022A ELISA with wavelength at 570 nm to calculate the inhibitory rate of proliferation. The growth density and morphologic change of Lewis lung cancer cells were observed by inversion microscope. Results Methyl mercury could inhibit the proliferation of rats Lewis lung cancer cells, and this effect had time and dose-dependence. The difference of the same concentration among the different time and that of the same time among the different concentrations were significant ( P 〈 0. 001 ). The inhibition of methyl mercury on the proliferation of Lewis cells of rats in lung cancer was stronger than that of As2O3 and cisplatin. The effect concentration and speed of methyl mercury was lower and quicker than that of As2O3 and cisplatin. Conclusions Methyl mercury could inhibit the proliferation of rats Lewis lung cancer cells, its effect has time and dose-dependence, and this effect is stronger than As2O3 's and cisplatin's.
