J61321(Alteromonas aurantia)菌株产铁载体培养条件的研究 被引量:5

摘要 在摇瓶水平上采用单次单因子方法对铁载体高产菌株J61321(Alteromonas au- rantia)的生长和产铁载体的发酵条件进行优化,考察了碳源、氮源、培养时间、培养温度、初始pH值、摇床转速等因素的影响,获得菌株J61321产铁载体的最佳发酵条件:酵母粉0.1%(W/V),蛋白胨0.2%(W/V),FePO_40.01g·L^(-1),温度28℃,转速180 r·min^(-1), pH7.5,铁载体在菌株J61321培养22 h后达到最大产量。在最适培养条件下测定了出发菌株A18和突变株J61321不同培养时间的菌液OD_(600nm)值、铁载体产量和抑菌活性,结果显示二者的生长量差距不大(p>0.05),但是铁载体产量和抑菌活性之间存在显著性差异(p<0.05)。 The optimal medium components (carbon and nitrogen sources) and growth conditions (initial pH, temperature and rotation speed) for siderphore production by mutant J61321 (Alteromonas aurantia ) were studied by using one-variable-at-a-time method. The results showed that the optimal medium components for the highest yield of siderphore production by mutant J61321 were 0. 1% (W/V) yeast extraction, 0.2% (W/V) peptone, 0.01 g · L^-1 FePO4, and the growth conditions for the highest yield of siderphore production by mutant J61321 were 180 r · min^-1 rotation speed, initial pH= 7.5, temperature 28 ℃. Under this condition, the siderphore production reached the highest yield after 22 h of fermentation in the liquid medium. The biomass, siderophore yield and antibacterial activities of the mutant J61321 were compared with thf parent A18. The results showed that the biomass of the two strains has no difference (p〉 0.05) during the growth, whereas the siderophore yield and antibacterial activities from the mutant J61321 were higher than that of the parent strain (p〈0.05).
