摘要
目的探索不同浓度表皮生长因子(EGF)对人脐血间充质干细胞(Mesenchymal stem cells,MSCs)增殖的影响。方法无菌条件下来集正常人脐血,密度梯度离心法分离脐血单个核细胞,用Mesencult培养基进行纯化和扩增培养,通过MTT法检测不同浓度的EGF的促人脐血MSCs增殖作用。结果EGF的5—100ng/ml各浓度组增殖效应均明显大于对照组(P〈0.05),10ng/ml组明显大于其它各浓度组(P〈0.01)。结论EGF对体外培养的人脐血MSCs有促进增殖的作用,此作用呈剂量依赖性。
Objective To study the effect of epidermis growth factor (EGF) at different concentration on proliferation of human umbilical cord mesenchymal stem cells. Methods Human cord blood samples were obtained sterilely and stored in preservativefree heparin. Density gradient eentrifugation methods were used for isolation of the cord blood mononuelear cells. Plating the cells with Mesencuh medium, the influence of different doses of EGF on proliferation of MSCs was observed by MTT test. Results The enhanced effect in every concentration group of EGF (5ng/ml to 100ng/ml) was significantly stronger than that of the controls ( P 〈 0.05), the group with concentration of 10ng/mL was more significantly stronger than other groups ( P 〈 0.01 ). Conclusion EGF possesses the enhancement effect on the proliferation of MSCs in a depended manner.
