动脉粥样硬化兔血管内皮功能与黏附分子VCAM-1表达关系的研究 被引量:9

摘要 目的探讨高分辨超声检测动脉粥样硬化(AS)兔内皮功能与黏附分子VCAM-1表达的关系。方法日本大耳白兔27只,对照组(A组)6只,动脉粥样硬化模型组(B组)21只,各随机分为3个亚组,分别喂养4、8和12周。超声检测兔血管内皮依赖性舒张功能(EDD)和血管内皮非依赖性舒张功能(NEDD),并同时应用免疫组化方法检测了VCAM-1表达。结果B1组脂纹期EDD减低,VCAM-1表达量最多,主要表达于内皮细胞表面。在B2组进展期病变中,EDD进一步减低,整个组织内均有VCAM-1表达。在B3组粥样斑块时,EDD更进一步减低,同时NEDD也出现损伤,VCAM-1主要以平滑肌细胞表达为主,VCAM-1在正常状态下呈低表达。结论EDD损伤与内皮细胞VCAM-1的表达有关,而NEDD损伤与平滑肌细胞VCAM-1表达有关。 [Objective] To study the relationship between the vascular endothelium function and VCAM-1 in atherosclerotic rabbits during different feeding time. [Methods] Twenty seven Japanese white big rabbits were fed with normal feed (control group) or fed with cholesterol diet for 4.8 and 12 weeks. EDD (endothelial dependent dilation) and NEDD (non-endothelial dependent dilation) were measured with high resolution ultrasound. The expression of VCAM-1 in EIA were checked with immunohistochemical staining technique. [Results] In B1 group and VCAM- 1 expressed maily on the surface of endothelial cells. In B2 group EDD decreased progressively and VCAM-1 expressed in the whole tissue. In B3 group EDD and NEDD all decreased and VCAM-1 expressed in smooth muscle cells. [Conclusion] EDD damage is related to the expression of VCAM-1 on the endothelial cells. NEDD damage is related to the expression of VCAM-1 in the smooth muscle cells.
