摘要
目的建立血浆甲硝唑浓度的高效液相色谱(HPLC)测定方法。方法采用HPLC-UV方法,以法舒地尔为内标;色谱柱为Hypersil C18柱(5μm,4.6mm×200mm);流速:1.0mL·min^-1;柱温:30℃;流动相:0.02mol·L^-1磷酸二氢钠溶液(用1mol·L^-1磷酸溶液调节pH值=3.00):甲醇=80:20,紫外检测波长319nm。结果甲硝唑的血药浓度在0.25-20.00mg·L^-1范围内与峰面积比有良好的线性关系,定量下限为0.25mg·L^-1。方法的平均回收率为(98.24±1.30)%,日内、日间相对标准差(RSD)均〈10%。结论该方法简单、方便、准确,是血浆甲硝唑浓度检测较好的分析方法。
Objective To establish a HPLC method for determination of the serum metronidazole concentration. Methods A HPLC-UV assay was developed. The interstandard was fasudil. The determinate wavelength was 319nm. The colum was Hypersil C18 (5 μm, 4. 6 mm × 200 mm). The mobile phase was KH2PO4-methnol (80 : 20, V/V) , and the column temperature was 30℃. The flow rate was 1.0 mL·min^-1. Results The determination limit was 0. 25 mg·L^-1. The metmnidazole was linear in the range from 0. 25 mg·L^-1 to 20.0 mg·L^-1. The RSD was no more than 10.0%. Conclusion The HPLC method is simple, convenient and accurate, which provides a reference method for determination of serum metronidazole concentration.
