摘要
目的研究大鼠胚胎后肾间充质干细胞(MMSCs)体外诱导分化的生物学特性。方法原代分离培养后肾间充质干细胞,在体外特定诱导条件下观察细胞向成骨细胞、脂肪细胞及肾小管上皮细胞的分化及其形态演变,采用细胞免疫荧光检测细胞表面特定蛋白的表达变化。结果大鼠后肾间充质干细胞在特定诱导条件下可向成骨细胞、脂肪细胞及上皮细胞分化,免疫荧光结果显示分化后的上皮细胞E-cadherin蛋白表达明显增强,而vimentin、fibronectin的表达则明显降低甚至无表达。结论大鼠胚胎后肾间充质干细胞在特定条件下有定向分化的能力,具有潜在分化为肾实质细胞的能力。
Objective To investigate the biological characters of metanephric mesenchymal stem cells(MMSCs) derived from embryonic in differentiation. Methods Primary metanephric mesenchyme stem cells were cultivated and multi-directional differentiation was induced. Morphological changes were observed and the expression of vimentin,fibronectin and E-cadherin was measured by immunofluorescence technique. Results The MMSCs could be inuced to differentiate to adipocytes, osteocytes and epithelial cells in special conditions. Immunofluorescence results showed the expression of E-cadherin increased, but expression of vimentin and fibronectin decreased in the surfaces of differentiated renal epithelial cells. Conclusion Metanephric mesenchymal stem cells can be inuced to differentiate in specific directions and have the potent capability of differentiating to renal cells.
