非小细胞肺癌FasL异常表达与肿瘤浸润性淋巴细胞凋亡的关系 被引量:2

Relationship between up-regulation of Fas ligand expression and apoptosis of tumor infiltrating lymphocytes in human non-small cell lung cancer

摘要 目的探讨非小细胞肺癌FasL表达与肿瘤浸润淋巴细胞凋亡之间的关系。方法采用免疫组织化学方法和末端脱氧核苷酸转移酶介导核苷酸缺口标记(TUNEL)技术对45例非小细胞肺癌(NSCLC)FasL表达和肿瘤浸润淋巴细胞凋亡进行检测。结果非小细胞肺癌FasL阳性表达率(65.89±17.02)%。受检45例NSCLC组织肿瘤浸润淋巴细胞凋亡指数为4.69±1.97。TIL凋亡指数与肿瘤病理类型及分化程度有关,腺癌组织中TIL凋亡指数高于鳞癌(P<0.01);低分化肺癌TIL凋亡指数高于高分化肺癌(P<0.01);NSCLC组织中TIL凋亡指数与TNM分期及是否伴淋巴结转移无关(P>0.05)。TIL凋亡指数随NSCLC FasL表达强度增强而增加,与其表达强度呈显著正相关(r=0.707,P<0.01)。结论NSCLC表达FasL为肿瘤反杀伤宿主肿瘤浸润性T淋巴细胞提供了有利条件;NSCLC异常表达FasL和TIL细胞凋亡异常在肿瘤逃避宿主免疫监视、对机体免疫系统实施反杀伤方面具有非常重要的生物学意义。 Objective To explore the association between FasL expression and apoptosis of tumor infiltrating lymphocytes in human non-small cell lung cancer(NSCLC). Methods The expressions of FasL were detected in 45 cases of human non-small cell lung cancer by SP immunohistochemical technique and TdT-mediated-dUTP-X nick end labeling (TUNEL). Results (1) The positive FasL expression existed in all tested NSCLC tissues was (65.89±17.02)% in average. The apoptosis index (AI) of tumor infiltrating T lymphocytes (TIL) in the tested NSCLC tissues was 4.69 ± 1.97. AI of TIL had relationship with the phathological type and differontiation degree of the tumor: AI in adenocarcinoma was higher than that in squamous cell lung cancer ( P〈0. 01 ) AI in low differentiated NSCLC was significantly higher than that in high differentiated NSCLC(P〈0.01). AI had no association with TNM stage and lymph node metastasis or not. AI was growing with the increasing degree of FasL in NSCLC, with significant and positive corelationship (r=0. 707, P〈0.01). Conclusion FasL expression in NSCLC may be the advantageous condition that the tumor counterattacks tumor infiltrating T lymphcytes. Abnormal expression of FasL and apoptosis of tumor infiltrating lymphocytes in NSCLC may play very important biological roles in escaping the host immunosurveillance and enforcing counterattack to the host immunity.
