摘要
目的:分析猪囊尾蚴特异性抗原编码cDNA的阶段特异性表达机制。方法:用猪囊尾蚴特异性抗原编码cDNA(λcC28)及抗λcC28单表位抗体作探针,采用Northernblot、Southernblot和Westernblot分子杂交技术分析了λcC28编码cDNA在猪带绦虫各发育阶段的特异性表达。结果:猪囊尾蚴和成虫不同节片抗原蛋白具有阶段特异性,λcC28cDNA编码的28kDa抗原蛋白只在囊尾蚴阶段表达,成虫幼节、成节、孕节都不表达。结论:猪囊尾蚴阶段特异性抗原的编码基因存在于囊尾蚴阶段和成虫的幼节中,但仅有囊尾蚴阶段的mRNA表达28kDa抗原蛋白。
AIM:To examine the cDNA expression of the stage specific antigen in Cysticercus cellulosae. METHOD: The λcC28 gene expression was demonstrated by λcC28 cDNA and anti λcC28 monospecific antibody as probes by using Northern blot and Southern blot analysis. RESULTS: The 28 kDa antigenic protein was demonstrated only in Cysticercus, but not in any proglottids of adult worms. CONCLUSION: The λcC28 gene was transcripted only in Cysticercus , although this gene was demonstrated also in immature proglottids. Expression of λcC28 gene was controlled at transcription level.
