在垂体腺瘤中TSP-1和TGF-β1的表达及其与微血管密度的关系 被引量:6

Expressions of TSP-1 and TGF-β1 in pituitary adenoma and the associations to microvessei density

摘要 目的探讨凝血酶敏感蛋白1(TSP-1)和转化生长因子B1(TGF-β1)在垂体瘤中的表达以及与微血管密度(MVD)、侵袭性的关系。方法取52例垂体瘤(侵袭性组24例,非侵袭性组28例)手术标本,采用Western印迹方法检测TSP-1、TGF-β1蛋白表达,并分析其表达与垂体瘤侵袭性、MVD等因素之问的关系。MVD以CD34为指标采用免疫组化方法进行检测。结果与非侵袭组相比,侵袭组TSP-1蛋白表达较弱(0.435±0.052vs0.642±0.043,P〈0.01),TGF-β1蛋白表达则较强(0.533±0.061vs0.364±0.049,P〈0.01),两组间MVD表达差异无统计学意义。TSP-1表达与MVD无明显相关性,TGF-B1与侵袭性及MVD呈正相关(r=0.51,P〈0.05)。结论TSP-1和TGF-β1在垂体瘤发生发展中起重要作用,并影响其血管生成及侵袭性。 Objective To investigate the expressions of thrombospondin-1 (TSP-1) and transforming growth faetor-β1 (TGF-β1) in pituitary adenomas and their associations with invasiveness and microvessel density (MVD) of adenomas. Methods The expressions of TSP-1 and TGF-β1 in 52 surgical specimens (24 invasive eases and 28 non-invasive cases)of pituitary adenomas were measured by Western blot methods and the association of the expressions with MVD and invasiveness of adenomas was studied. MVD is measured by immunohistochemical method of CD34. Results Compared with the non-invasive group, the expression of TSP-1 was lower (0.435 ± 0.052 vs 0. 642 ± 0. 043 ) and the expression of TGF-β1 was higher ( 0. 533 ± 0. 061 vs 0. 364 ± 0. 049) in invasive group ( both P 〈0.01 ) , but expression of CD34 ( t = 2. 257, P = 0. 083 ) was similar in 2 groups. The expression of TGF-β1 had a positive correlations with MVD ( r = 0.51, P 〈 0.05 ), but there was no correlations between expression of CD34 and the invasion of pituitary adenomas. In addition, MVD was not associated with the expression of TSP-1. Conclnsion TSP-1 and TGF-β1 play an important role in the pathogenesis of pituitary adenomas and influence the invasiveness and angiogenesis.
