摘要 正常造血干细胞(HSC)具有自我更新和多向分化潜能,能在体内重建造血系统。20世纪70年代早期,一些研究表明白血病细胞中仅有一小群细胞有体外广泛增殖能力,基于这些研究,出现了白血病干细胞(leukemia stem cells, LSC)的概念:白血病细胞中极少数具有无限自我更新和起始白血病形成能力的细胞。体外培养技术、非肥胖糖尿病/严重联合免疫缺陷(NOD/SCID)鼠等移植模型以及免疫单抗表型、流式细胞术等实验技术的进展,使LSC的分离、鉴定成为可能,并最终在急性髓系白血病(AML)(除外M3)、急性淋巴细胞白血病(ALL)和慢性粒细胞白血病(CML)等疾病中都证明了LSC的存在。目前普遍认为,与正常造血过程类似,白血病的发生及维持都依赖于LSC。
