赤芍总苷对D-氨基半乳糖胺所致小鼠肝损伤的保护作用 被引量:14

摘要 目的观察赤芍总苷对D-氨基半乳糖胺(D-GalN)所致小鼠肝损伤的防治作用。方法NIH小鼠60只,随机分5组,分别为正常对照组,模型组,赤芍总苷高剂量组、低剂量组和联苯双脂组小鼠分别灌胃给药,1次/d,连续7d,末次给药1h后,除正常对照组外,其余各组小鼠均腹腔注射D-GalN 800mg/kg,16h后采血,检测血清ALT、AST活性及超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量,并观察肝组织病理学变化。结果赤芍总苷高剂量组能显著降低D-GalN所致小鼠肝损伤血清ALT、AST活性,提高肝组织匀浆SOD活性,降低MDA,明显改善肝组织损伤的程度,与模型组比较差异有统计学意义(P<0.05)。结论赤芍总苷对D-GalN诱导的小鼠肝损伤有保护作用,其作用机制可能与抗脂质过氧化有关。 Objective To study the protective effects of total paeony glycoside(TPG) on liver injury in mice induced by D-galactosamine. Method Sixty NIH mice were randomly divided into five groups: nomal control group (group A) ,model group (group B) ,high dosage group of TPG (group C) ,small dosage group of TPG (group D)and bifendate group (group E). Mice in group C, group D and group E were administered with drugs by gastric perfusion continue for 7 days, once daily. One hour after the last administration of the drugs, except those in group A, mice in other groups were injected with D-GaiN 800 mg/kg into its abdominal cavity. Blood samples were collected at 16 hour after D-GaIN administration and measured the levels of ALT, AST, superoxide dismutase (SOD) activity and malondiadehyde(MDA). Histopathological examination was also performed for liver tissue. Result High dosage TPG could significantly lower the levels of serum ALT, AST and MDA in the liver tissue, but enhanced SOD activity in the liver tissue and markedly ameliorated the liver damage (P 〈 0.05). Conclusion TPG can protect against acute liver injury induced by D-GaiN in mice. The mechanism of which maybe related to its antiperoxidation of lipid.
