肿瘤坏死因子的细胞同步化作用对鼻咽癌放射敏感性的影响 被引量:1

摘要 目的探讨肿瘤坏死因子(TNFα)使细胞周期同步化对增加鼻咽癌的放射敏感性的影响。方法用体内实验证实鼻咽癌细胞系中确实存在放射抵抗亚克隆株,然后用流式细胞仪检测TNFα对CNE-2Z-S1细胞周期同步化的作用,应用克隆形成实验及裸鼠实验,于体外体内测定经细胞周期同步化后S1细胞对射线的敏感性。结果(1)CNE-2Z细胞群中确实存在着放射抵抗的亚群。(2)TNFα能阻滞CNE-2Z-S1细胞周期,达到细胞周期同步化的作用。(3)体外研究证实经TNFα预处理可使CNE-2Z-S1细胞对放射的敏感性显著提高。(4)体内研究提示TNFα有可能提高CNE-2Z-S1细胞对放射的敏感性。结论TNFα可通过促进CNE-2Z-S1细胞周期同步化进而增加放射敏感性。 Objective To investigate the effect of tumor necrosis factor α(TNFα) on radiosensitivity of nasopharyngeal carcinoma (NPC) in relation to TNFa-induced cell cycle synchronization. Methods The radio-resistance ofa NPC cell line subclone CNE-2Z-S1 was verified by in vivo experiments and flow cytometry was performed to evaluate cell cycle synchronization in TNFα-treated CNE-2Z-S1 cells. The radiosensitivity of the cell synchronized CNE-2Z-S1 cells was determined by clone formation vitro and in vivo experiment in nude mice. Results TNFα was capable of inducing cell cycle arrest and synchronization of CNE-2Z-S1 cells. Pretreatment with TNFα remarkably enhanced the radiosensitivity of CNE-2Z-S1/n vitro, and in vivo experiments with nude mice also suggested the role of TNFct in enhancing the radiosensitivity ofNPC. Conclusion TNFα can enhance the radiosensitivity of NPC ceils by inducing cell cycle synchronization.
