亚砷酸钠对血红素加氧酶-1诱导作用 被引量:1

摘要 目的观察不同暴露剂量和时间下,亚砷酸钠(NaAsO2)对体外培养的牛主动脉血管内皮细胞(BAEC)中血红素加氧酶-1(HO-1)的诱导作用。方法分别在0-15μmol/L NaAsO2暴露0-24 h收集培养的BAEC细胞蛋白提取液中,用蛋白印迹(western blot)法检测HO-1蛋白的诱导产生量。结果0-15μmol/L NaAsO2暴露能够显著诱导BAEC细胞HO-1蛋白产生增加,并且分别具有剂量-反应和时间-反应相关关系;2.5μmol/L NaAsO2暴露即可显著诱导产生HO-1蛋白。结论HO-1蛋白的诱导表明砷介导的一种细胞应激反应;HO-1蛋白可以作为无机砷暴露的敏感性生物标志物之一,有利于深入研究无机砷暴露的生物剂量反应。 Objective To observe sodium arsenite (NaAsO2) -induced expression of heme oxygenase- 1 (HO- 1)in bovine aortic endothelial cells (BAEC), Methods Cultured BAEC was exposed to NaASO2 (0- 15μmol/L) for 0- 24 h, respectively. Western blot was used to detect the induction of HO- 1 protein. Results 0 15 μmol/L NaAsO2 exposure induced the production of HO - 1 protein significantly, with dose - dependent and time - dependent relationship, Low dose of NaAsO2(2.5 μmol/L) could induce markedly the expression of HO - 1 protein, Conclusion HO - 1 induction could be a cellular stress response, and has potential for being used as a sensitive biomarker for inorganic arsenic exposure, which may be useful for further research of the biological response of arsenic.
