摘要
以COL-0型野生型拟南芥菜为材料,使用枪头在1.5ml的Eppendorf管中直接捣碎的方式,用Trizol试剂提取总RNA。通过总RNA的纯度、浓度测定及琼脂糖凝胶电泳分析提取效果。测定结果表明样品纯度分别达到1.9843和1.9741,浓度分别为1.2061μg/μl和1.1187μg/μl,电泳结果显示总RNA的28SrRNA、18SrRNA和5SrRNA三条带清晰、无拖尾。与研钵研磨材料比较,此方法简便、需要材料少、降低了提取过程中RNA的降解几率。
Arabidopsis thahana total RNA was isolated with Trizol reagent, by gun tip directly pounding to pieces in 1.Sml Eppendorf tube. The result shows that the purity of each sample is 1.9843 and 1.9741; Concentration 1.2061 μg/μ1 and 1.1187 μg/μl, and agar gel electrophoresis shows that the three bands of 28S rRNA, 18S rRNA and 5S rRNA are clear. Compared with pounding to pieces by mortar, this method is simple, needing less material and reducing the probability of RNA fall.
