福辛普利拉对培养乳鼠心肌细胞缺氧/复氧损伤的保护作用

摘要 目的探讨福辛普利拉对培养乳鼠心肌细胞缺氧/复氧损伤的保护作用。方法取SD新生大鼠(1~3d)120只,培养成心肌细胞,在培养的第4d随机分为5组(每次取20只,每组重复6次):正常对照组、缺氧/复氧(A/R)组、福辛普利拉剂量1+A/R(F1+A/R)组、福辛普利拉剂量2+A/R(F2+A/R)组、福辛普利拉剂量3+A/R(F3+A/R)组。建立缺氧/复氧损伤模型,以福辛普利拉进行干预,对各组分别观察心肌细胞搏动频率、细胞存活率和培养液中乳酸脱氢酶活性。结果①细胞搏动频率(beat/min):A/R组(11.50±3.39)比正常对照组(99.83±4.22)明显减低(P〈0.01);福辛普利拉各剂量组比A/R组显著加快(P〈0.01),比正常对照组稍慢,但无显著性差异(P〉0.05)。②细胞存活率:A/R(26.78±4.25%)组比正常对照组(87.96±2.58)明显降低(P〈0.01);福辛普利拉各剂量组比A/R组明显增高(P〈0.01),与正常对照组相比无显著性差异(P〉0.05)。③LDH活性:A/R组(69.50±7.34)比正常对照组(10.33±2.73)明显升高(P〈0.01);福辛普利拉各剂量组比A/R组明显降低(P〈0.01),与正常对照组相比虽有升高但无显著性差异(P〉0.05)。结论从细胞水平证实福辛普利拉预处理对心肌细胞对缺氧/复氧损伤具有保护作用。 Objective To investigate the mechanisms of anoxia/reoxygenation and the effects of Fosinoprilat on the cultured cardiocytes. Methods Cultured cardiocytes of neonatal SD rats (1-3 d)were randomly divided into five groups: control group; anoxia reoxygenation (A/R) group; Fosinoprilatl (F1 + A/R) group; Fosinoprilat2 (F2 + A/R) group and Fosinoprilat3 (F3 + A/R) group. In each group, the cardiocytes were subjected to anoxia (3 hr) followed by reoxygenation (1 hr) on the model of the cultured myocardial cells. At the end of experiment, cardiocytes beating rate,cell viability and the activity of LDH in culture were measured. Results (1). Beating rate of cardiocytes: The beating rate was significantly slower in A/R group(11. 50±3.39beat/min) compared with that of control group(99.83±4.22 beat/min)(P〈〈0.01);Between each group of Fosinoprilat and control group, the beating rate was similar to each other(P〉0. 05) ; Compared with A/R , the beating rate of each group of Fosinoprilat was significantly increased(P〈0.01).(2). the cell viability: The cell viability of A/R(26.78±4.25) was significantly decreased when compared with that of control group(87.96 ±2.58 %)(P〈0. 01) ; The cell viability of each group of Fosinoprilat significantly increased when compared with A/R(P〈0. 01); Between each group of Fosinoprilat and control group, the cell viability was similar to each other(P〉0.05). (3). LDH activity: LDH activity of A/R (69.50±7.34 IU/L) was significantly increased when compared with control group(10.33±2.73)(P〈0.01) ; LDH activity of each group of Fosinoprilat significantly increased when compared with A/R(P〈0. 01); Between each group of Fosinoprilat and control group, the LDH activity was similar to each other(P〉0. 05). Conclusion It is suggested that Fosinoprilat may be involved in the process of protection on cultured cardiocytes in anoxia/ reoxvygenation iniurv.
