一种经济简单的方法扩增Vγ9Vδ2T细胞

One economical and simple method to amplify Vγ9Vδ2T cells

摘要 目的体外以仲丁胺刺激健康人外周血PBMC中Vγ9Vδ2T细胞的增殖,为γδT细胞的过继免疫治疗建立效应细胞制备平台。方法淋巴细胞分离液分离健康人外周血的PBMC,以10^6/孔接种于24孔板,用含10%胎牛血清、1mmol/L仲丁烷和200UIL-2的1640完全培基连续培养2周。结果有效地扩增出了Vγ9Vδ2T细胞,其组成高达85%以上,所扩增出的Vγ9Vδ2T细胞能有效地杀伤Daudi细胞。结论使用小分子的胺基抗原能有效地扩增出有功能的Vγ9Vδ2T细胞,为γδT细胞的过继免疫治疗建立了效应细胞制备平台。 Objective To amplify enough Vγ9Vδ2T cells for the adoptive therapy of tumor. Methods 106 PBMC, isolated from healthy human peripheral blood with Ficoll, were seeded in 24-well plate and cultured in 1640 medium supplied with 10% fetal bovine serum, 1 mmol/L sec-butylamine and 200 U IL-2 for two weeks. Results The method could effectively promote Vγ9Vδ2T cells to proliferate. Two weeks culture produced enough Vγ9Vδ2T cells which took up to 85% of the total PBMC and displayed powerful eytotoxieity to Daudi cells. Conlusion Small molecule sec-butylamine could effectively amplify functional Vγ9Vδ2T cells, which provided enough effector cells for the adoptive therapy of tumor.
